शट रुजा-विशरण-गत्यवसादनेषु शटति शटी, अचि ङीः| केचित् षटीति मूर्धन्यादिमाहुः, शढीत्यपि, तस्याम्| पलाशाः सन्त्यस्य पलाशः, अभ्रादित्वादः| षड् ग्रन्थयोऽस्याः षड्ग्रन्था| गन्धयति-अर्दयति गन्धोली, “पिञ्छोल-कल्लोल-” [हैमोणादिसू. ४९५] इत्योले निपात्यते| हिमे जाता हिमजा| उह्यते वधूः, “वहेर्ध् च” [हैमोणादिसू. ८२२] इत्यूः| करोति कर्चूरः, “सिन्दूर-कर्चूर-पत्तूर-धत्तूरादयः” [हैमोणादिसू. ४३०] इत्यूरे निपात्यते| शोभनो(?नं) व्रतो(?तम)ऽस्याः सुव्रता| गन्धयुक्तं मूलमस्याः गन्धमूली| पृथ्वी चासौ पलाशिका च पृथुपलाशिका| आह च–

कृन्तति त्वग्दोषं कर्चूरः, सिन्दूर-कर्चूर-पत्तूर-धत्तूरादयः [हैमोणादिसू. ४३०] इत्यूरे निपात्यते, तत्र| द्रावयति द्राविडः, विहड-कहोड- [हैमोणादिसू. १७२] इत्यडे निपात्यते| “कल्यकारी काल्यः” इति क्षीरस्वामी| वेधे अग्र्यः-श्रेष्ठो वेधाग्र्यः| गन्धयुक्तं मूलमस्य गन्धमूलः, गन्धो मूले अस्येति वा, के गन्धमूलकः| आह च–

| 1 | Pharmacological evaluation of extracts of Hedychium spicatum (Ham-ex-Smith) rhizome | Hedychium spicatum (Ham-ex-Smith), known as Shati in Ayurvedic classics, is documented for the treatment of cough, hiccough, fever and asthma. The present study includes the evaluation of aqueous and ethanolic extracts of the dried rhizome of H. spicatum for anti-histaminic and ulcer-protective activities in guinea pig (GP), anti-inflammatory and analgesic activities in rat and acute toxicity in mouse. The extracts were administered orally, daily as suspension, in 1% carboxymethyl cellulose either for 7 days in GP studies or 60 min before or just before experiment in rats and mice. An initial dose-dependent anti-histaminic action of both the extracts (100, 200 and 400 mg/kg) was performed against histamine-induced bronchospasm in GPs. The 200 mg/ kg dose of aqueous and ethanolic extracts was selected both in GP and rat for further studies. GPs treated with aqueous and ethanolic extracts showed gastric ulcer protection against histamine-induced gastric ulcer compared with the control group. Both the extracts also showed an anti-inflammatory effect against carrageenan-induced paw edema in rats from 1 h onwards, and this was maximum at 3 h. Analgesic effect was determined by using hot plate and tail flick tests in rats, and both the extracts at 200 mg/kg showed a significant increase in the latent period from 30 min onwards till 120 min of their study period. Both the extracts did not show any toxic effect like increased motor activity, salivation, clonic convulsion, coma and death in mice even at the 2000 mg/kg dose (nearly 10 times of the optimal effective dose), indicating the safety of the extracts. Don in west Himalaya, India | Total phenolic compounds and antioxidant potential of Hedychium spicatum were analyzed for 16 different natural populations located in Uttarakhand (west Himalaya) for promotion as health or medicinal food. Total phenolic compounds varied among populations from 4.70 mg gallic acid equivalent (GAE) to 2.84 mg GAE/g dry weight. Three in vitro antioxidant assays, i.e. azinobisethylbenzothiazoline-6-sulphonic acid radical scavenging (ABTS) assay, diphenyl-2-picrylhydrazyl (DPPH) assay and ferric reducing antioxidant power (FRAP) assay, showed significant (p < 0.05) differences across populations. Antioxidant activity and phenolic contents of the methanolic extract of the leaves and rhizomes of Hedychium spicatum were evaluated using in vitro standard methods. Spectrophotometry was the bases of the determination of the total tannin, total phenol, total flavonoids, total flavonols and total proanthocyanidins, tannic acid, gallic acid, rutin, catechin equivalents were used for these parameter. The antioxidant activity of methanolic extract of the leaves and rhizomes of Hedychium spicatum was determined by DPPH radical scavenging assay. The results from this study showed that the antioxidant activity of leaves and rhizomes of Hedychium spicatum as determined by the total tannin, phenolics, flavonoids, flavonols and proanthocyanidins level present. The leaves extract has high free radical scavenging activity as shown in 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay than the rhizomes methanolic extract. |
| 8 | Hedychium spicatum rhizome essential oil induces apoptosis in human prostate adenocarcinoma PC-3 cells via mitochondrial stress and caspase activation | Hedychium spicatum is an essential oil bearing plant extensively used in the traditional system of medicine in several countries. Previous research has revealed H. spicatum essential oil (HSEO) to exhibit anti-tumor activity, although the mechanism of action is still unknown. Therefore, the current study was designed to carry out a comprehensive characterization of HSEO and evaluate the chemotherapeutic potential of HSEO against cancerous cells. The volatile constituents of HSEO was determined by one-dimensional gas chromatography with time-of-flight mass spectrometry (GC-TOFMS) and two-dimensional gas chromatography with time-of-flight mass spectrometry (GCxGC-TOFMS). In total, 193 phytocompounds could be detected, out of which 140 were identified for the first time. The major phytoconstituents detected by GCxGC-TOFMS were β-pinene (10.94%), eucalyptol (6.45%), sabinene (5.48%) and trans-isolimonene (5.00%). GCxGC-TOFMS analysis showed two and half fold increase in the constituents over GC-TOFMS due to better chromatographic separation of constituents in the 2nd dimensional column. HSEO was tested for in vitro cytotoxic activity against cancerous (PC-3, HCT-116 and A-549) and normal (3T3-L1) cell, with HSEO being most selective for prostate cancer cell (PC-3) over non-tumorigenic fibroblast (3T3-L1) cell. HSEO treatment inhibited the colony formation ability of PC-3 cells. HSEO treatment caused apoptotic cell death and cell cycle arrest at G2/M and S phase in PC-3 cells. HSEO induced apoptosis via intracellular ROS accumulation, mitochondria depolarization and increased caspase-3, 8, and 9 levels in PC-3 cells. Additionally, HSEO treatment led to a decrease of Bcl-2 and Bcl-xL and increase of Bax and Bak protein levels. Overall, results from this study highlighted the anticancer potential of H. spicatum essential oil, which could be considered as a new agent for treating prostate cancer. |
| 9 | Comparative study on eucalyptol and camphor rich essential oils from rhizomes of Hedychium spicatum Sm. and their pharmacological, antioxidant and antifungal activities | The present communication reveals the variation in essential oil composition of Hedychium spicatum collected from different altitudinal locations viz., Nainital, Uttarakhand and Renuka, Himachal Pradesh in India. Both the oils were found to be rich in oxygenated sesquiterpenes, hydrogenated sesquiterpenes and oxygenated diterpenes. The essential oils exhibited antifungal activity which might be possibly due to thepresence of several phytoconstituents. Thus, it can be concluded that oil of this important herb can be used for the development of natural fungicides, after proper screening and clinical studies. The ability of Hedychium spicatum to reduce the volume of paw edema validates the essential oil’s ability to regulate the inflammatory mechanisms, and thus helping in the natural treatment of inflammatory swelling, ulcers, sores, glandular swelling, wounds and cuts. The essential oils of H. spicatum are also able to reduce the oxidative stress by scavenging the reactive oxygen species like free radicals, hydroxy free radicals, superoxide free radicals, alkoxy free radicals and non-radicals like hydrogen peroxides etc and can be helpful to cure various pathological conditions. Based on these findings, the herb Hedychium spicatum being a rich source of biologically active oxygenated terpenoids, can be utilised for the development of natural antioxidants, anti-inflammatory, anti-nociceptive and anti-pyretic drugs. Besides, the generation of evidence-based database for future research and judicious in-situ exploitation of the herb. |
